Antonia San Juan was born in Las Palmas de Gran Canaria, Spain. When she was 19, she moved to Madrid. There, she began her career as an actress in plays. She also performed in cabaret shows in different places.

She became famous for her role as Agrado in the movie All About My Mother (Todo sobre mi madre). This film was directed by the well-known filmmaker Pedro Almodóvar. In Spain, she is well-known not just for her movies. She is also famous for her funny solo performances, called monologues, on TV and in theaters.

Since 2009, Antonia San Juan has been part of the popular Spanish TV series La que se avecina. In this show, she plays the character of Estela Reynolds. Estela is the funny and unique mother of Lola.
