Clarksville is in the northeastern part of Greene County, Pennsylvania. It sits where two creeks, Tenmile Creek and its South Fork, meet.

The town is surrounded by other areas. To the west is Morgan Township. To the south and east, across the South Fork Tenmile Creek, is Jefferson Township. To the north, across Tenmile Creek, is East Bethlehem Township in Washington County.
Clarksville is about 3 miles (4.8 km) southwest of Millsboro. This is where Tenmile Creek flows into the Monongahela River. Sometimes, a nearby part of East Bethlehem Township is also thought of as part of Clarksville.
