Nicki Micheaux was born in Detroit, Michigan. Her father was an officer in the United States Army. She went to the University of Colorado Boulder for college.

She decided to take an acting class because she thought it would be an easy grade. However, she quickly discovered a passion for performing. When she acted, she felt a huge rush of excitement that would last for days. The university often used race-blind casting, which means they chose the best actor for the role, no matter their race. Micheaux started her career with small parts in movies and guest appearances on TV shows. Some of her early appearances were on popular shows like ER and Any Day Now.

From 2001 to 2002, she had a recurring role on the drama series Soul Food. She also appeared in the popular shows Six Feet Under and The Shield. In 2005, she acted alongside Halle Berry in the TV movie Their Eyes Were Watching God.
