In 1501, Antoine became a chamberlain for Philip the Handsome. A chamberlain was a high-ranking official who managed the royal household.
Later, in 1510, he became a Counsellor and Chamberlain for the young Charles of Luxemburg. Charles later became the Holy Roman Emperor, one of the most powerful leaders in Europe.

Antoine was also a general. He played a big part in helping the Burgundian dukes fight against Charles, Duke of Guelders. Charles was trying to take over the Sticht area. Antoine was also a strong Catholic. He worked hard to stop the spread of Protestantism, which was a new religious movement at the time. He even ordered the building of the Sint-Catharinakerk (Saint Catherine's Church) in Hoogstraten.
In 1530, Antoine became the acting governor of the Netherlands. This happened because Archduchess Margaret of Austria, the main governor, was having health problems. In Antoine's last year of life, Charles V appointed his sister, Mary of Austria, to take over as governor. Antoine de Lalaing passed away in 1540.
