Guilford is in the eastern part of Franklin County, Pennsylvania. Its exact location is 39°55′10″N 77°35′54″W / 39.91944°N 77.59833°W. Most of Guilford is in Guilford Township, Pennsylvania. A small part of it also reaches north into Greene Township. People sometimes call this area Guilford Hills.

A major road, U.S. Route 30, goes through Guilford. This road can take you about 2 miles (3.2 km) east to Fayetteville. If you go west for about 4 miles (6.4 km), you will reach Chambersburg. Chambersburg is the main town, or county seat, of Franklin County.
