After college, Mike started working as a sports reporter and anchor at WMAQ-AM radio in Chicago. He later moved to WSCR-Radio, where he covered big events like the World Series and the Super Bowl. From 1993 to 1995, he also wrote a weekly sports column for newspapers. In 1994, he started reporting for SportsChannel Chicago. By 1995, he was an anchor and host for CLTV in Chicago. In 1999, Mike Greenberg was asked to co-host a new morning radio show on ESPN Radio with Mike Golic. He agreed, but he also kept working on SportsCenter. Their show, Mike & Mike, became very popular. In 2004, the show started being shown on TV on ESPNEWS. Because it was so successful, it moved to ESPN2 in 2005.

One fun part of Mike & Mike was the yearly "Sheet of Integrity" bet. This was a friendly competition based on the NCAA men's basketball tournament. Mike Golic would enter many different prediction sheets, but Mike Greenberg believed in only using one "Sheet of Integrity." The loser of the bet had to do something funny on the air.

For example, one year Mike Greenberg won, and Mike Golic had to get his eyebrow waxed on TV! Another time, Mike Golic won, and Mike Greenberg had to wear the University of Notre Dame Leprechaun mascot costume. In 2007, Mike Greenberg, who is a big New York Jets fan, agreed to wear a New England Patriots jersey to a Jets game. He also had to milk a cow live on the show!

Mike Greenberg's last day as co-host of Mike & Mike was November 17, 2017. Both Mike Greenberg and Mike Golic were honored for their work. They were put into the National Association of Broadcasters’ Broadcasting Hall of Fame in 2016. In 2018, they joined the National Radio Hall of Fame. After Mike & Mike ended, Mike Greenberg started hosting a new ESPN morning show called Get Up! in April 2018. He also returned to ESPN Radio with his new show, Greeny.
