East Petersburg is located in the northern part of Lancaster County. It's about 5 miles (8 km) northwest of Lancaster city. You can find it at these coordinates: 40°6′0″N 76°21′10″W / 40.10000°N 76.35278°W.
The main road through town is Pennsylvania Route 72. This road connects East Petersburg to nearby towns like Manheim and Lancaster. Another important road, Pennsylvania Route 722, crosses Main Street in the center of town.
