Penryn is in the northern part of Lancaster County. It's about 3.5 miles northeast of a town called Manheim. It is also about 6 miles northwest of Lititz. The county seat, Lancaster, is about 13 miles south of Penryn.

Most of this area is land, with only a very small part, about 0.01 square miles (0.03 square kilometers), being water. The community gets its water from small streams that flow into Chiques Creek. This creek then flows south directly into the Susquehanna River.
