De Kalb was originally known as Bloomington, and under the latter name was laid out in 1839. It was renamed in 1851 for Johann von Robais, Baron de Kalb, a major general in the American Revolutionary War. A post office called De Kalb has been in operation since 1840.

According to the United States Census Bureau, the city has a total area of 0.25 square miles (0.65 km2), all of it land.
As of the census of 2010, there were 220 people, 87 households, and 65 families residing in the city. The population density was 880.0 inhabitants per square mile (339.8/km2). There were 97 housing units at an average density of 388.0 per square mile (149.8/km2). The racial makeup of the city was 99.1% White, 0.5% African American, and 0.5% from two or more races. Hispanic or Latino of any race were 0.5% of the population.
There were 87 households of which 40.2% had children under the age of 18 living with them, 59.8% were married couples living together, 11.5% had a female householder with no husband present, 3.4% had a male householder with no wife present, and 25.3% were non-families. 18.4% of all households were made up of individuals and 12.6% had someone living alone who was 65 years of age or older. The average household size was 2.53 and the average family size was 2.86.
The median age in the city was 37.3 years. 25.5% of residents were under the age of 18; 10.5% were between the ages of 18 and 24; 26% were from 25 to 44; 23.2% were from 45 to 64; and 15% were 65 years of age or older. The gender makeup of the city was 47.7% male and 52.3% female.
As of the census of 2000, there were 257 people, 101 households, and 73 families residing in the town. The population density was 1,017.0 people per square mile (396.9/km²). In the town the population was spread out with 25.7% under the age of 18, 8.2% from 18 to 24, 31.5% from 25 to 44, 19.1% from 45 to 64, and 15.6% who were 65 years of age or older. The median age was 34 years. For every 100 females there were 97.7 males. For every 100 females age 18 and over, there were 101.1 males.
