Before Europeans settled here, Native American tribes lived in the area. These included the Quapaw and later the Cherokee people. Some famous figures from the Wild West, like Jim Bowie and Jesse James, traveled through the Greenbrier region. They were often heading towards Oklahoma and Texas.

Around 1818, the Wiley brothers settled near East Fork Cadron. This spot was about 8 miles (13 km) east of where Greenbrier is today. Jonathan Hardin settled near the Wiley family in 1837. Also, in the early 1800s, the Casharagos family settled north of Greenbrier. This is now the community of Republican.

The town got its name from the greenbriar vines. These vines grew all along the creek that runs through the town. Greenbrier is about 12 miles (19 km) north of Conway. Many people who work in Conway now choose to live in Greenbrier. This has made Greenbrier a growing "bedroom community".
