Before playing professionally, Kwamie played college football for the Kansas Jayhawks. In 2022, he joined the Cincinnati Bengals as a player who was not chosen in the main draft.

But he quickly rejoined them on their practice squad the next day. On November 26, 2022, he was moved to the active roster. This allowed him to play in a game against the Tennessee Titans.

On January 31, 2023, Kwamie signed a new contract with the Bengals. He was released again on August 29, 2023. The very next day, he signed back onto their practice squad. On October 7, 2023, he was moved to the active roster for a game. This game was against the Arizona Cardinals. It was only his second NFL game ever. His father, Kwamie Lassiter, played for the Cardinals for eight seasons. In this game, Kwamie II made his first professional catch. It was a 2-yard reception. After the game, he returned to the practice squad.

The Bengals signed Kwamie to another contract on January 8, 2024. However, he was released by the team on August 23, 2024.
