Barkers Creek is a small river, or a tributary, in southern West Virginia, United States. It is about 8 miles (13 km) long. This creek flows into the larger Guyandotte River.

Through the Guyandotte and Ohio rivers, Barkers Creek is part of a huge watershed. This means all the water from its area eventually flows into the Mississippi River. The creek drains an area of about 37 square miles (96 km²). This area is a rural part of the Allegheny Plateau, which was not shaped by glaciers.

Barkers Creek and all the land it drains are located in southeastern Wyoming County. The creek starts in the very eastern part of Wyoming County. This is near where Wyoming, Raleigh, and Mercer counties meet. It begins about 3 miles (5 km) northeast of a place called Arista.

First, Barkers Creek flows west. Then, it turns and flows northwest. It passes through small communities like Bud, Alpoca, and Tralee. The creek joins the Guyandotte River from the south. This meeting point is about 0.5 miles (0.8 km) northwest of Tralee. It is also about 1 mile (1.6 km) southeast of Itmann. A road called West Virginia Route 10 runs alongside the lower part of the creek.
