Parnell started his professional cricket career at just 17 years old. He played his first first-class cricket match for Eastern Province in October 2006. In only his fifth first-class match, he achieved a rare feat: he took a hat-trick against Western Province. This means he got three wickets in three balls! People quickly started talking about him as a very promising young cricketer.

In 2008, Wayne won awards for being the best under-19 player in South Africa. In February 2022, he was chosen to be the captain of the Western Province team for the 2021–22 CSA T20 Challenge tournament.
