Several important roads pass through Quarryville. U.S. Route 222 goes through the borough. It leads northwest about 14 miles to Lancaster, which is the main city of the county. If you go south on Route 222 for about 17 miles, you'll reach U.S. Route 1 in Conowingo, Maryland.
Pennsylvania Route 372, also known as State Street, also runs through Quarryville. It goes east about 16 miles to Parkesburg. If you head west on Route 372 for about 11 miles, you'll cross the Susquehanna River near Holtwood. Another road, Pennsylvania Route 472 (South Lime Street), leads southeast from Quarryville about 13 miles to Oxford.

The town got its name from a stone quarry that was next to the village. Many workers lived in this village because of the quarry. As more workers came, the village grew bigger, and that's how it became known as "Quarryville." People who live there say that one day, the workers went to lunch and came back to find the quarry half-full of water. The quarry site is still there today, filled with water.
