Osorio began his professional career with SC Toronto in 2012. This team played in the Canadian Soccer League. He had a great first season, scoring many goals. He was the club's second-highest goal scorer.

His team finished third in the league, which earned them a spot in the playoffs. They lost their playoff game against the Serbian White Eagles FC. Because of his strong first year, Osorio was named the CSL Rookie of the Year.

Playing for Toronto FC

Jonathan Osorio joined Toronto FC's main team in 2013. He had been training with their youth academy since 2012. He impressed the coach, Ryan Nelsen, during preseason training. This led to him getting a contract with the first team.

He played his first game for Toronto FC on March 9, 2013. He scored his first goal on March 30, 2013, against the Los Angeles Galaxy. His second goal came on April 27 against the New York Red Bulls. This goal was even named the MLS Goal of the Week. After the 2013 season, Osorio trained with teams in England and Germany.

In the 2017 season, Osorio played less often at the start. This was because another player, Víctor Vázquez, joined the team. However, Osorio started in important playoff games. He played in the 2017 MLS Cup final, which Toronto FC won 2-0 against Seattle Sounders FC.

Osorio had a very strong start to the 2018 season. Teams from Europe and Mexico showed interest in him. He won the Golden Boot award in the 2018 CONCACAF Champions League. He scored 4 goals in that tournament.

In the 2018 Canadian Championship final, Osorio scored a goal in the first game. In the second game, he helped set up a goal. This was his 200th game for Toronto FC. Toronto FC won the championship, and Osorio was named the most valuable player. He signed a new contract with Toronto FC in August 2018. This made him one of the highest-paid Canadian soccer players.

On November 10, 2019, Osorio played in the 2019 MLS Cup Final. Toronto FC lost 3-1 to Seattle Sounders. In April 2022, Osorio played his 300th game for Toronto FC. He was the first player to reach this milestone for the club.

His contract with Toronto FC ended after the 2022 season. However, in December 2022, Toronto FC announced that Osorio had signed a new three-year contract. This contract also included an option for 2026. In March 2023, it was shared that his new contract made him a "Designated Player." This means his salary does not count fully against the team's salary cap. Before the 2024 season, he was named the club captain.
